RESUMO

Animals are inherently motivated to explore social novelty cues over familiar ones, resulting in a novelty preference (NP), although the behavioral and circuit bases underlying NP are unclear. Combining calcium and neurotransmitter sensors with fiber photometry and optogenetics in mice, we find that mesolimbic dopamine (DA) neurotransmission is strongly and predominantly activated by social novelty controlling bout length of interaction during NP, a response significantly reduced by familiarity. In contrast, interpeduncular nucleus (IPN) GABAergic neurons that project to the lateral dorsal tegmentum (LDTg) were inhibited by social novelty but activated during terminations with familiar social stimuli. Inhibition of this pathway during NP increased interaction and bout length with familiar social stimuli, while activation reduced interaction and bout length with novel social stimuli via decreasing DA neurotransmission. These data indicate interest towards novel social stimuli is encoded by mesolimbic DA which is dynamically regulated by an IPN→LDTg circuit to control NP.

RESUMO

Pituitary neuroendocrine tumors (PitNETs) account for approximately 15% of all intracranial neoplasms. Although they usually appear to be benign, some tumors display worse behavior, displaying rapid growth, invasion, refractoriness to treatment, and recurrence. Increasing evidence supports the role of primary cilia (PC) in regulating cancer development. Here, we showed that PC are significantly increased in PitNETs and are associated with increased tumor invasion and recurrence. Serial electron micrographs of PITNETs demonstrated different ciliation phenotypes (dot-like versus normal-like cilia) that represented PC at different stages of ciliogenesis. Molecular findings demonstrated that 123 ciliary-associated genes (eg, doublecortin domain containing protein 2, Sintaxin-3, and centriolar coiled-coil protein 110) were dysregulated in PitNETs, representing the upregulation of markers at different stages of intracellular ciliogenesis. Our results demonstrate, for the first time, that ciliogenesis is increased in PitNETs, suggesting that this process might be used as a potential target for therapy in the future.

The structure of isolated 1-chloronaphthalene has been investigated in a supersonic expansion by high-resolution chirped-pulse Fourier transform microwave (CP-FTMW) spectroscopy in the 2-8 GHz frequency range. Accurate values of the rotational, centrifugal distortion, and nuclear quadrupole coupling constants for the only availabe conformer have been determined. The intensity of the spectrum allowed us to observe all the heavy atoms isotopologues in natural abundance, determining their rotational constants. From the extensive experimental dataset we derived accurate structures for 1-chloronaphthalene using different methodologies and compared with related compounds.

Objective: To assess the clinical impact of flash glucose monitoring (FGM) systems on fear of hypoglycemia (FoH) and quality of life in adults with type 1 diabetes mellitus (T1DM). Methods: Prospective quasi-experimental study with a 12-month follow-up. People with T1DM (18-80 years old) and self-monitoring by blood capillary glycemia controls were included. The FH15 questionnaire, a survey validated in Spanish in a comparable study population, was used to diagnose FoH with a cutoff point of 28 points. Results: A total of 181 participants were included, with a FoH prevalence of 69% (n = 123). A mean reduction in FH15 score of -4 points (95% confidence interval [-5.5 to -3]; P < 0.001) was observed, along with an improvement in quality of life (EsDQOL-test (Diabetes Quality of Life, Spanish version), -7 points [-10; -4], P < 0.001) and satisfaction with treatment (Diabetes Treatment Satisfaction questionnaire, self-reported version [DTSQ-s] test, +4.5 points [4; 5.5], P < 0.001). At the end of the follow-up, 64.2% of the participants saw an improved FoH intensity, compared to 35.8% who scored the same or higher. This improvement in FoH status was associated with a higher time-in-range at the end of the follow-up (P = 0.003), as well as a lower time spent in hyperglycemia (P = 0.005). In addition, it was linked to participants with a high baseline FoH levels (P < 0.001) and those who were university degree holders (P = 0.07). Conclusions: FGM is associated with an overall reduction of FoH in adults with T1DM and with an increase in their quality of life. Nevertheless, a significant percentage of patients may experience an increase of this phenomenon leading to clinical repercussions and a profound impact on quality of life.

The aim of the present study was to analyze the sporting progression from U14 to senior categories of elite Spanish high and long jumpers. For prospective analysis, 300 athletes ranked top 20 at U14 were analyzed (153 female and 147 male). For retrospective analysis, 64 athletes ranked in the top 20 in the senior category were included (21 female and 43 male). Ranking positions were registered in each of the seasons where they presented records. Only 6.3% (19) of athletes who reached the top 20 at U14 became successful senior athletes [4.7% (14) of athletes maintained top 20 status throughout their sporting careers from U14 to senior]. The transition rate from U14 to U16 (35.7%) was the most severe drop down in consecutive categories (after this, it ranged from 47.8 to 66.7%). Of the senior top 20 athletes (64), most of them were already ranked top 20 at U16 (59.4%, 38), at U18 (62.5%, 40), at U20 (70.3%, 45) and at U23 (78.1%, 50). Nevertheless, only 34.4% (22) were top 20 when they were U14 athletes. Transition rates in the top 20 senior athletes ranged from 86.7 to 95.5%. Around one out of four (26.6%, 17) of the top 20 senior athletes maintained top 20 status throughout their sporting careers from U14 to the senior category. Although early success is not a good predictor of senior success, successful senior athletes excelled early on and were able to remain in top rankings throughout their sporting careers of national elite jumpers.

BACKGROUND: Most total disc replacements provide excessive mobility and not reproduce spinal kinematics, inducing zygapophyseal joint arthritic changes and chronic back pain. In cadaveric lumbosacral spines, we studied if a new lumbar disc prosthesis kinematics mimics the intact intervertebral disc. METHODS: In eight cold preserved cadaveric lumbosacral spines, we registered the movement ranges in flexion, extension, right and left lateral bending, and rotation in the intact status, post-discectomy, and after our prosthesis implantation, comparing them for each specimen. FINDINGS: Comparing the intact lumbosacral spine with the L4-L5 prosthesis implanted specimens, we saw statistically significant differences in lateral bending and right rotation but not in the full range of rotation. Analyzing segments, we also noticed statistically significant differences at L4-L5 in flexion-extension and rotation. On the other hand, the L4-L5 discectomy, compared to the baseline spine condition, showed a statistically significant mobility increase in flexion, extension, lateral bending, and axial rotation, with an abnormal instantaneous center of rotation, which destabilizes the segment partly due to anterior annulus surgical removal. Disc prosthesis implantation reversed these changes in instantaneous center of rotation, but the prosthesis failed to restore the initial range of motion due to the destabilization of the ligaments in the operated disc. INTERPRETATION: The ADDISC total disc replacement reproduces the intact disc kinematics and Instantaneous Center of Rotation, but the prosthesis fails to restore the initial range of motion due to ligament destabilization. More studies will be necessary to define a technique that restores the damaged ligaments when implanting the prosthesis.

CONTEXT: Treatment with Alemtuzumab (ALZ) in patients with Relapsing-Remitting Multiple Sclerosis (RRMS) is associated with the development of ALZ-induced Graves' disease (GD-ALZ). Some cases may develop associated Graves´ Orbitopathy (GO-ALZ), with possible visual compromise. AIM: The aim of this study was to describe the main clinical and biochemical characteristics of GD-ALZ, as well as the clinical course of a case series of GO-ALZ METHODS: This study is a retrospective observational study, carried out in a reference hospital for the care of patients with RRMS in Spain. Cases treated with ALZ in the period 2014-2022 were included. GO-ALZ cases were identified among those with clinical symptoms compatible with thyroid eye disease after initiating ALZ treatment. RESULTS: A total of 135 cases, with a mean follow-up of 69.6 months after the first ALZ cycle, were included. The incidence of GD-ALZ was 32.6% (44/135), with a predominance of women (77.3%) and mean age of 41.9 years. The presence of first-degree relatives with hypothyroidism was identified as risk factor for the development of GD-ALZ (adjusted P-value: 0.02). GO-ALZ was diagnosed in 6 cases (incidence: 13.6%), of which 3 had severe clinical forms of GO, requiring anti-IL-6 treatment. A favorable response was reported in all of them, with a significant decrease in disease activity and improvement in proptosis. CONCLUSIONS: We report one of the largest cohorts of GD-ALZ and GO-ALZ cases. The diagnosis of these entities should be taken into account in patients treated with Alemtuzumab, given the risk of developing severe clinical forms. In moderate-severe forms of GO-ALZ, drugs with anti-IL-6 activity are a safe and effective option.

Previous research has shown that, unlike misspelled common words, misspelled brand names are sensitive to visual letter similarity effects (e.g., amazom is often recognized as a legitimate brand name, but not amazot). This pattern poses problems for those models that assume that word identification is exclusively based on abstract codes. Here, we investigated the role of visual letter similarity using another type of word often presented in a more homogenous format than common words: city names. We found a visual letter similarity effect for misspelled city names (e.g., Barcetona was often recognized as a word, but not Barcesona) for relatively short durations of the stimuli (200 ms; Experiment 2), but not when the stimuli were presented until response (Experiment 1). Notably, misspelled common words did not show a visual letter similarity effect for brief 200- and 150-ms durations (e.g., votume was not as often recognized as a word than vosume; Experiments 3-4). These findings provide further evidence that the consistency in the format of presentations may shape the representation of words in the mental lexicon, which may be more salient in scenarios where processing resources are limited (e.g., brief exposure presentations).

BACKGROUND: Recently, vesicles derived from plant cell membranes have received attention for their potential use as active biomolecules and nanocarriers, and obtaining them from organic crops may be an interesting option because different farming systems can affect production, plant secondary metabolism and biochemistry of cell membranes. The present study aimed to determine how organic and conventional farming affects the mineral nutrition, gas exchange, CO2 fixation and biochemical composition of lemon fruits, which could have an impact on the different fractions of cell membranes in pulp and juice. RESULTS: Organic trees had higher intrinsic water use efficiency (WUEi) but conventional trees had higher stomatal conductance (gs) and nitrogen use efficiency (NUtE). Also, organic lemons had significantly higher levels of some micronutrients (Ca, Cu, Fe and Zn). Second, the main differences in the membrane vesicles showed that organic pulp vesicles had a higher antioxidant activity and more oleic acid, whereas both types of vesicles from conventional lemons had more linoleic acid. CONCLUSION: In conclusion, organic farming did not alter carbon fixation parameters but impacted nitrogen fixation and water uptake, and resulted in higher micronutrient levels in lemons. These mineral nutritional changes could be related to the higher production of membranes that showed suitable morphological traits and a high antioxidant activity, positively correlated with a high amount of oleic acid, which could have stronger cell protection characteristics. © 2023 The Authors. This paper focuses on the experimental characterization of the polarization behavior of light backscattered through fog. A polarimetric orthogonal state contrast imager and an active, purely polarized white illuminator system are used to evaluate both linear and circular polarization signals. The experiments are carried out in a macro-scale fog chamber under controlled artificial fog conditions. We explore the effect of backscattering in each imaging channel, and the persistence of both polarization signals as a function of meteorological visibility. We confirm the presence of the polarization memory effect with circularly polarized light, and, as a consequence, the maintenance of helicity in backscattering. Moreover, the circular cross-polarized channel is found to be the imaging channel less affected by fog backscattering. These results are useful and should be taken into account when considering active polarimetric imaging techniques for outdoor applications under foggy conditions.

The increase in the reptile population has led to a rise in the number of zoonotic infections due to close contact with reptiles, with reptile-associated salmonellosis being particularly relevant. California kingsnake invasion not only threatens the endemic reptile population of the island of Gran Canaria (Spain) but also poses serious public health problems by spreading zoonotic pathogens and their antimicrobial resistance (AMR) to the environment. Thus, the aim of this study was to assess the occurrence, genetic diversity, and AMR among Salmonella spp. strains isolated from California kingsnakes in Gran Canaria Island (Spain). Of 73 invasive individuals captured, 20.5% carried Salmonella spp., belonging to different subspecies and serovars, with subsp. salamae as the most abundant. Pulsed-field electrophoresis showed high genetic diversity among subsp. salamae isolates, and among these, 73.3% showed resistance to at least one of the antimicrobials tested. In conclusion, the present study revealed the importance of wild invasive California kingsnakes as reservoirs of drug-resistant Salmonella spp. that could pose a direct threat to livestock and humans. Identification of drug-resistant Salmonella strains in wildlife provides valuable information on potential routes of transmission that involve risks to public and animal health.

Cardiovascular diseases cause a high number of deaths nowadays. To improve these statistics, new strategies to better understand the electrical and mechanical abnormalities underlying them are urgently required. This study focuses on the development of a sensor to measure tissue stretch in excised tissues, enabling improved knowledge of biomechanical properties and allowing greater control in real time. A system made of biocompatible materials is described, which is based on two cantilevered platforms that integrate an optical fiber inside them to quantify the amount of stretch the tissues are exposed to with a precision of µm. The operating principle of the sensor is based on the variation of the optical path with the movement of the platforms onto which the samples are fixed. The conducted tests highlight that this system, based on a simple topology and technology, is capable of achieving the desired purpose (a resolution of ∼1 µm), enabling the tissue to be bathed in any medium within the system.

Multiligamentary knee injuries associated with transtibial amputation is a pathologie with a low incidence, so their diagnosis and treatment represent a great challenge for the medical team, mainly due to the low rate of scientific publications on the matter. This article intends to present the treatment of a really infrequent pathology, presenting the clinical case of a polytraumatized patient who suffered a left transtibial amputation associated with a multiligament knee injury with dislocation of the proximal tibiofibular joint. After analyzing the multiple therapeutic options, a specific surgical planning is carried out for the specific case of a multiligament knee injury associated with an ipsilateral transtibial amputation, proceeding to the execution of the reconstruction of the injury with good results. For this, several specific surgical gestures are carried out, adapted to the patient's condition, which will facilitate the surgery and are explained in the surgical technique. In conclusion, we must know that in order to obtain satisfactory results in these patients, it is important to carry out an early diagnosis and treatment of the injury, analyzing the proximal tibiofibular stability and providing adequate stability through the execution of a precise surgical technique.

RESUMO

Peer review is a core component of scientific practice. Although peer review ideally improves research and promotes rigor, it also has consequences for what types of research are published and cited and who wants to (and is able to) advance in research-focused careers. Despite these consequences, few reviewers or editors receive training or oversight to ensure their feedback is helpful, professional, and culturally sensitive. Here, we critically examine the peer-review system in psychology and neuroscience at multiple levels, from ideas to institutions, interactions, and individuals. We highlight initiatives that aim to change the normative negativity of peer review and provide authors with constructive, actionable feedback that is sensitive to diverse identities, methods, topics, and environments. We conclude with a call to action for how individuals, groups, and organizations can improve the culture of peer review. We provide examples of how changes in the peer-review system can be made with an eye to diversity (increasing the range of identities and experiences constituting the field), equity (fair processes and outcomes across groups), and inclusion (experiences that promote belonging across groups). These changes can improve scientists' experience of peer review, promote diverse perspectives and identities, and enhance the quality and impact of science. To investigate whether local elements are grouped into global shapes in the absence of awareness, we introduced two different masked priming designs (e.g., the classic dissociation paradigm and a trial-wise probe and prime discrimination task) and collected both objective (i.e., performance based) and subjective (using the perceptual awareness scale [PAS]) awareness measures. Prime visibility was manipulated using three different prime-mask stimulus onset asynchronies (SOAs) and an unmasked condition. Our results showed that assessing prime visibility trial-wise heavily interfered with masked priming preventing any prime facilitation effect. The implementation of Bayesian regression models, which predict priming effects for participants whose awareness levels are at chance level, provided strong evidence in favor of the hypothesis that local elements group into global shape in the absence of awareness for SOAs longer than 50 ms, suggesting that prime-mask SOA is a crucial factor in the processing of the global shape without awareness.

This manuscript addresses the phenomenon of masked priming and the cognitive process of switching from Spanish to English while reading in sequential bilingual texts compared to heritage speakers. A lexical decision task was employed in the present study with masked translation priming, which serves as a valuable tool for elucidating the orthographic and lexical processes involved in the initial stages of reading. This study builds upon previous research conducted on monolingual masked priming, which consistently demonstrates shifts in the response time (RT) distributions when comparing related and unrelated primes. Within the framework of a diffusion model, we implemented two theoretical positions. First, we posited that translation priming operates at the orthographic level, resulting in enhanced efficiency during the encoding process. Second, we explored the possibility that translation priming operates at the semantic level, influencing the accumulation of evidence during the lexical decision task. The findings of the present study indicate that translation priming elicits outcomes similar to those observed in monolingual priming paradigms. Specifically, we observed that translation priming facilitation is manifested as shifts in the RT distributions. These findings are interpreted to suggest that the benefits derived from the encoding process are not specific to the accessed lexicon following a brief stimulus presentation.

PURPOSE: to perform an external validation of our predictive model to rule out pheochromocytoma (PHEO) based on unenhanced CT in a cohort of patients with PHEOs and adenomas who underwent adrenalectomy. METHODS: The predictive model was previously developed in a retrospective cohort of 1131 patients presenting with adrenal lesions. In the present study, we performed an external validation of the model in another cohort of 214 patients with available histopathological results. RESULTS: For the external validation, 115 patients with PHEOs and 99 with adenomas were included. Our previously described predictive model combining the variables of high lipid content and tumor size in unenhanced CT (AUC-ROC: 0.961) had a lower diagnostic accuracy in our current study population for the prediction of PHEO (AUC: 0.750). However, when we excluded atypical adenomas (with Hounsfield units (HU) > 10, n = 39), the diagnostic accuracy increased to 87.4%. In addition, in the whole cohort (including atypical adenomas), when MRI information was included in the model, the diagnostic accuracy increased to up to 85% when the variables tumor size, high lipid content in an unenhanced CT scan, and hyperintensity in the T2 sequence in MRI were included. The probability of PHEO was <0.3% for adrenal lesions <20 mm with >10 HU and without hyperintensity in T2. CONCLUSION: Our study confirms that our predictive model combining tumor size and lipid content has high reliability for the prediction of PHEO when atypical adrenal lesions are excluded. However, for atypical adrenal lesions with >10 HU in an unenhanced CT scan, MRI information is necessary for a proper exclusion of the PHEO diagnosis.

The sense of touch is underrepresented in cognitive psychology research. One of the reasons is that controlling the timing of stimulus presentation, which is a hallmark of cognitive research, is significantly more difficult for tactile stimuli than visual or auditory stimuli. In the present work, we present a system to display tactile stimuli (braille cells) and collect response time with the capability for static and dynamic (passive haptic) stimuli prsentation that will contribute to the development of tactile research. While the system requires some construction, it can be put together with commercially available materials. Here, we present the step-by-step instructions for constructing the tool, the code used to control it, and some basic experiments to validate it. The data from the experiments show that the device can be used for a variety of tactile perception experiments.

BACKGROUND AND PURPOSE: Alcohol abuse has been associated with erectile dysfunction (ED), but the implicated molecular mechanisms are unresolved. This study analyses the role of alterations in soluble guanylyl cyclase (sGC) in ED. EXPERIMENTAL APPROACH: ED was analysed in adult male C57BL/6J mice subjected to the Chronic Intermittent Ethanol (CIE) paradigm. Erectile function was assessed in anaesthetised mice in vivo by evaluating intracavernosal pressure (ICP) and in vitro in isolated mice corpora cavernosa (CC) mounted in a myograph. Protein expression and reactive oxygen species were analysed by western blot and dihydroethidium staining, respectively. KEY RESULTS: In CIE mice, we observed a significant decrease in the relaxant response of the CC to stimulation of NO release from nitrergic nerves by electrical field stimulation, to NO release from endothelial cells by acetylcholine, to the PDE5 inhibitor sildenafil, and to the sGC stimulator riociguat. Conversely, the response to the sGC activator cinaciguat, whose action is independent of the oxidation state of sGC, was significantly enhanced in these CC. The responses to adenylyl cyclase stimulation with forskolin were unchanged. We found an increase in reactive oxygen species in the CC from CIE mice as well as an increase in CYP2E1 and NOX2 protein expression. In vivo pre-treatment with tempol prevented alcohol-induced erectile dysfunction. CONCLUSIONS AND IMPLICATIONS: Our results demonstrate that alcoholic mice show ED in vitro and in vivo due to an alteration in the redox state of sGC and suggest that sGC activators may be effective in ED associated with alcoholism.

BACKGROUND: Intervertebral disc nucleus degeneration initiates a degenerative cascade and can induce chronic low back pain. Nucleus replacement aims to replace the nucleus while the annulus is still intact. Over time, several designs have been introduced, but the definitive solution continues to be elusive. Therefore, we aimed to create a new nucleus replacement that replicates intact intervertebral disc biomechanics, and thus has the potential for clinical applications. MATERIALS AND METHODS: Two implants with an outer ring and one (D2) with an additional midline strut were compared. Static and fatigue tests were performed with an INSTRON 8874 following the American Society for Testing and Materials F2267-04, F2346-05, 2077-03, D2990-01, and WK4863. Implant stiffness was analyzed at 0-300 N, 500-2000 N, and 2000-6000 N and implant compression at 300 N, 1000 N, 2000 N, and 6000 N. Wear tests were performed following ISO 18192-1:2008 and 18192-2:2010. GNU Octave software was used to calculate movement angles and parameters. The statistical analysis package R was used with the Deducer user interface. Statistically significant differences between the two designs were analyzed with ANOVA, followed by a post hoc analysis. RESULTS: D1 had better behavior in unconfined compression tests, while D2 showed a "jump." D2 deformed 1 mm more than D1. Sterilized implants were more rigid and deformed less. Both designs showed similar behavior under confined compression and when adding shear. A silicone annulus minimized differences between the designs. Wear under compression fatigue was negligible for D1 but permanent for D2. D1 suffered permanent height deformation but kept its width. D2 suffered less height loss than D1 but underwent a permanent width deformation. Both designs showed excellent responses to compression fatigue with no breaks, cracks, or delamination. At 10 million cycles, D2 showed 3-times higher wear than D1. D1 had better and more homogeneous behavior, and its wear was relatively low. It showed good mechanical endurance under dynamic loading conditions, with excellent response to axial compression fatigue loading without functional failure after long-term testing. CONCLUSION: D1 performed better than D2. Further studies in cadaveric specimens, and eventually in a clinical setting, are recommended. Level of evidence 2c.
